If someone has already booked a Superliner standard bedroom, can a second person, with coach fare, stay in the room, or would they need to purchase the extra accomodations as well?

Yes - all that would need to happen is for the second person to have a coach fare ticket. You may want to see if a "Rail Sale" ticket is cheaper than coach fare through a regular Amtrak agent. The person who booked the standard room paid the full accommodation charge (which is for two people).

It's very possible that, even if there's not a rail sale going on, the ticket to add a second person to a sleeping car accommodation will come out cheaper than coach fare would have been. That's because the "rail fare" portion of a sleeping car trip is often cheaper than the coach fare between the same cities.
And by rebooking the second person's ticket to show that they're in a sleeping car, that opens up another coach seat for Amtrak to sell as well.
